Labelled diagram of Variegated leaf before after starch test: Variegated leaves are those that have different colors on their leaves. When these leaves are exposed to sunlight, they show different colors. Take a potted plant with variegated leaves – for example, money plant or croton.
Keep the plant in a dark room for three days so that all the starch is used up. Now keep the plant in the sun for about six hours. Remove a leaf from the plant. Mark on the green parts of leaves and trace it on a sheet of paper.
Dip the leaves in hot boiling water for a minute. After this, dip it in a beaker containing alcohol. Heat until the alcohol begins to boil. You will see the leaves change color. Now dip the leaves in a dilute solution of iodine for a few minutes.
Remove the leaf and rinse with iodine solution. Observe the leaf color and compare it with the initial leaf tracing. In this experiment we can prove that chlorophyll is necessary for the process of photosynthesis.
It has been found that the leaf is boiled and washed in alcohol and water before the starch test and it is yellow in color. After the starch test it is found that the leaf will turn blue-black.
